Investigation of 2’-formamidonucleoside modification on seed-based off-target effects of siRNA

Summary:

We are seeking a highly motivated intern to join our Medicinal Chemistry team, focused on the chemical synthesis of modified nucleosides and oligonucleotides. The intern will participate in synthesizing chemical building blocks for solid-phase oligonucleotide synthesis, as well as incorporation, purification and characterization of said entities. The ideal candidate will have a basic understanding of chemical synthesis, chemical biology in addition to the ability to work collaboratively in a fast-paced environment.

 

Intern Responsibilities: 

Nucleoside/ Oligonucleotide Synthesis: Synthesis, purification and characterization of modified phosphoramidite building blocks. Use of automated synthesizers to produce oligonucleotides. Purify oligonucleotides using various techniques such as HPLC, PAGE or other relevant methods. Research and Development Participate in development of siRNA/ oligonucleotide modalities Maintain accurate and detailed laboratory records in accordance with company policies and regulatory requirements. Communicate findings and progress to project leaders.

Requirements: 

Currently enrolled in a chemistry/ biochemistry undergraduate/ graduate program in the United States. Knowledge of basic organic chemistry terms, procedures and characterization. Hands on chemistry lab experience is a bonus. Excellent problem-solving and analytical skills Strong communication and interpersonal skills. Ability to work independently and collaboratively in a team environment. Must be able to being internship between 6/9 and 6/16/2025. Must be able and willing to remain in the program through at least 8/15/2025 Must be able and willing to commit to a minimum 40-hour workweek and work onsite in Carlsbad, CA. The student intern will be directly integrated into the Medicinal Chemistry team. This will include participating in weekly meetings where the student will occasionally present the progress of their research. In addition, the intern will participate in the chemical synthesis of nucleic acid building blocks using organic chemistry, followed by their characterization using spectroscopic techniques. These building blocks with be further incorporated in synthetic oligonucleotides for use in in vitro biological assays. The intern will be further responsible for proper laboratory practices including lab notebook upkeep and reagent preparation.
