Off-target effects present a significant challenge in the pharmaceutical industry, often resulting in adverse side effects and decreased therapeutic efficacy. This study is designed to evaluate two distinct structure-based drug design (SBDD) methods focused on addressing hERG liabilities within compound series and sub-series across various projects. Through comparative analysis, we can demonstrate the strengths and limitations of each method in mitigating associated side effects. The binding modes identified for a series of compounds could provide insights for modifying functional groups, potentially reducing off-target affinity. By refining these strategies with experimental validation, we aim to enhance the drug design process, leading to safer and more effective therapeutics. Ultimately, the selected SBDD workflow will be integrated with our machine learning models to create a comprehensive in silico strategy. **Skills:** **Familiarity with protein structures and biochemistry** **Comfort working in a Linux-based computing environment throughout the duration of the internship** **Excellent oral and written communication skills** **An open mind and willingness to learn** **Qualifications** **Requirement:** **Legally/permanently authorized to work in the US with no sponsorship required for employment visa status (e.g. H1-B status) now or in the future.** **M.S. or Ph.D. students pursuing studies in computational chemistry, cheminformatics, or a relevant field** **Currently enrolled in an accredited college or university** **Candidates must be available to work 40 hours/week** Permanently authorized to work in the U.S., must not require sponsorship of an employment visa (e.g., H-1B or green card) at the time of application or in the future.
