Il bioassay for discovery lab è un laboratorio dedicato allo sviluppo di saggi in vitro immunologici e funzionali finalizzati alla predizione dell’efficacia di vaccini e anticopri monoclonali diretti contro batteri e allo studio dell’interazione tra ospite e patogeno. Lo scopo ultimo di questo progetto è quello di sviluppare saggi immunologici per rilevare anticorpi specifici per gonococco in soggetti immunizzati con 4CmenB o con pregressa infezione da neissera gonorrhoeae. Lo stagista avrà altresì la possibilità di lavorare allo sviluppo di un saggio per misurare la risposta anticorpale specifica per chlamyida trachomatis in sieri animali o umani.   Cosa imparerai? Lo stage permetterà allo studente di acquisire competenze di immunologia, in particolare di serologia, finalizzate allo studio della risposta umorale sviluppata a seguito di infezione naturale o vaccinazione. Lo stagista avrà la possibilità di apprendere le seguenti tecniche: Sviluppo di saggi di binding di anticopi sia policlonali che monoclonal a batteri o antigeni batterici attraverso varie tecniche (ELISA, Luminex, citofluorimetria di flusso); Sviluppo di saggi funzionali (saggio di battericida, opsonofagocitosi, neutralizzazione, deposizione del complemento) per testare la capacità di anticorpi policlonali animali o umani o di anticorpi monoclonali di uccidere i batteri contro i quali sono stati indotti. Basi di statistica, analisi di correlazione tra risultati derivanti da saggi diversi. Estrazioni ed elaborazione dei dati prodotti tramite Excel, GraphPad e successiva presentazione dei dati in fromato power point.
