Lead Researcher in Blue Sky Protein Kinases
St. Jude Children's Research Hospital
We are looking for an experienced biochemist to participate in our exciting studies of the human protein kinome. As part of the largest BlueSky initiative, “Seeing the Invisible in Protein Kinases” at St. Jude Children's Researcher Hospital, we are expanding our structural studies of the human kinome to identify novel kinase-binding ligands. We are looking for experienced biochemist to aid in these efforts. The candidate will work in a fast-paced research environment, studying protein kinases by generating and characterizing high quality proteins, assess binding affinity and kinetics using various biophysical techniques in collaboration with NMR spectroscopists and computational biologists. This role will involve working closely as part of a multidisciplinary team including computational chemists, structural biologists, biochemists, and other biophysical specialists. The unparalleled NMR center at St. Jude includes three 600 MHz, two 700 MHz, six 800 MHz, 850 MHz, and the first 1.1 GHz in the world. Responsibilities: Performs tasks related to the research project independently, but within specific guidelines and subject to regular review by supervisor or other research staff. Perform basic molecular biology techniques like PCR, vector construction, plasmid purification and sequencing analysis. Responsible for expression, purification, modification, and characterization of recombinant proteins from a variety of host cells. Design and implement purification schemes and protein analytics, present results, and interpretation. Lead troubleshooting efforts for protein expression and purification processes. Work effectively in a collaborative team environment where results are achieved through influence, cooperation, and the incorporation of multiple points of view. Ideal Candidate: Proven expertise in advanced molecular cloning techniques. Experience generating chimeric recombinant sequences, site-directed mutagenesis, random mutagenesis, codon optimization, and/or gene synthesis is highly desired. Strong background in protein biochemistry, protein purification (using affinity and traditional chromatographic techniques), and biophysical characterization. Hands-on experience with instrumentation essential for protein purification and characterization, especially BioRad/AKTA FPLC, SDS PAGE and Western Blot. Work in an independent manner to perform protein analysis including (but not limited to) CE SDS, ITC, SPR, U/HPLC, DSF, and cell based functional assays. Excellent oral, presentation and written communication skills required. Minimum Education and/or Training: Bachelor’s degree in a relevant scientific area is required. Minimum Experience: 5+ years of research experience is required with a Bachelor's degree. 3+ years of experience is required with a Master’s degree. No prior experience required with a PhD. Strong knowledge and experience in understanding and application of underlying principles related to techniques being performed. Experience in R&D methods and processes in lab setting, and execution of research projects for the laboratory preferred. Proven performance in earlier role/comparable role.
